Of a well-known family, Gauis Julius Caesar was born on the 12th of 100BCE. He may have obtained his leadership skills at a very early age due to the fact that he derived from a political family. When Caesar s father died, he became the head of the family. This was the moment when Gauis Julius Caesar took a step forward to excel as the most powerful Roman general during his time. Two years has passed and Caesar has already found his second wife, Pompeia. She was the daughter of Quintus Pompeius Rufus the son of former consul and Cornelia(Caesar s deceased wife). But they were not m arried for long, Caesar divorced her for suspiciously being part of a crime on 63 BC. Four years later, Caesar was married to his last wife, Calpurnia. But they did not have any children. Death. The republic of Rome has survived for four hundred years. It s an empire without an emperor. Ruled by elected officials and written laws. The senate is at the very top of the society, an elite class of men. All equal, all desperately competing for fame and glory. Their ambition is the force that fuels the state. Discipline is not easy, in fact, it is hard, but the results are what we discipline ourselves for. Celebration of Discipline was a very eye opening and insightful book in many aspects. Techniques for Providing Fresh Water to Arid Regions Student ID Number: 36845 Introduction Water is vital as it does not have a substitute and is a finite resource (Kaplan, 2011). The demand for water is ever increasing but the supply is decreasing or is constant. This demand is brought up by factors such as population increase, industrialization and agricultural needs. Some areas receive the right amount of whereas, others do not receive the right amount. These regions are known as arid regions where rainfall and water supply is less yearly causing droughts. The report will examine the feasibility of two techniques namely desalination and building dams and reservoirs for supplying fresh water to arid regions considering factors such as cost, technology, location and water quality. Background Kaplan (2011) points out that there is approximately 36 million cubic kilometres of fresh water available on the earth which equals to 3% and the rest 97% is salt water. 77% of the fresh water available is locked up in the polar icecaps, glaciers and icebergs, less than 1% in fresh water lakes, springs and rivers. The rest is found in plants, atmosphere and human bodies. However, another argument put forward by Kaplan (2011) was that just over a half of the fresh water available on the earth is being used. This means that there is plenty of fresh water available but is inefficiently used, wasted in simpler terms and not evenly distributed across the world. Therefore, arid regions are formed as they do not receive the right amount of water demanded from their population in that area. UNEP (1992) reports about 47% of the surface of the earth being arid or semi-arid. This shows almost half of the world is without water whereas, some places have plenty of fresh water available. Another r eport of UNEP (1997) claims about 2 billion people live in arid regions and in many cases in very poor conditions. This shows the need and importance of development of ideas and techniques to provide fresh water to these regions. From the table; Africa and Asia have the largest extension of arid zones, they account for more than a half of worlds arid regions (FAO, 2002). Comparison of options: Desalination According to Kucera (2014), desalination is defined as the â€Å"process of removing dissolved solids, such as salts and minerals, from water.† Desalination was used practically first in the sixteenth and the seventeenth centuries by sailors. Desalination is sometimes referred as distillation and sailors back in the sixteenth century used simple distillation technique to evaporate sea water and collecting the fresh water. The figure below from Kucera (2014) shows the total desalination capacity by different countries since 2003. The highest percentages of desalination capacity are led by the Middle East countries i.e. Saudi Arabia and UAE followed closely by; Spain, USA, China, Algeria, Australia, Israel, and India, respectively. Figure 2 Kucera (2014) Figure 2 above also shows that desalination is a process that is mainly operated by the developing countries. Not a single African nation is seen in the figure means that the most arid region of the world is not using desalination process. Keyes , Conrad Fahy, Tansel, Berrin (2012) claim desalination have two major technological methods involved i.e. membrane and evaporative also known as thermal. Middle East use the evaporative technology while, the USA depend on the membrane process of desalination as it is more technologically advanced and has a cost advantage. â€Å"Desalination water treatment plants must be managed in an environmentally compatible manner and requires significant environmental assessment efforts depending on the quality of the concentrate and geographical location of the facility† (ibid, 2012). This shows the process of desalination for providing fresh water to arid region is quite costly and requires attention to the environment as only developing countr ies like the UAE and the USA can afford to use. Building dams reservoirs: A dam is a structure built along rivers or streams that are used to control the water flow and store the water in reservoirs during the rainy season and release as needed during the times of drought. Reservoirs are artificial lakes that store the water (UNEP, 2000). The stored water is used in several ways; irrigation, domestic use, producing hydro-electricity and leisure activities. .Abu-zeid and.El-Shibini (2010) states â€Å"the only main source for Egypt’s water is the River Nile.† Therefore Egypt is also considered part of the arid regions of the world. The Aswan High Dam was built on the River Nile in Egypt to control floods, produce HEP- Hydro-electric power and irrigate their lands in 1971. The dam had positive impacts on rapid growing population of Aswan province. The tourism industry and the fishers noted a growth in their businesses. The majority of the Egyptian population also depends on the river for its water for domestic and industrial use. The figure bel ow represents the area in hectares-ha of irrigation in countries of Africa in 1997. (University of Michigan, n.d.) The figure shows Egypt irrigated 3.3 million hectares in 1997, and that number will keep on increasing as population increases (University of Michigan, n.d). However, some negative impacts are noted by Abu-zeid and El-Shibini (2010) that during the construction of the dam several Egyptian residents and monuments had to immigrate to the cities of Lower Egypt. The dam being 111 metres high and 3830 metres long costs the Egyptian government about 1 billion dollars (Encyclopaedia Britannica, 2014). This shows building dams and reservoirs can have some drawbacks in the short run like cost, re-settlement, natural disasters and water borne diseases but in the long run it also improves the economy of the country and it helps the population of the country in irrigation, power and leisure activities. Recommendation and Conclusion It can be concluded that to provide water to arid regions of the world both the techniques are feasible. Desalination works when the region has a source of salt or seawater. Although, it is quite costly and not very friendly to the environment, it is now widely used in many countries which prove it to be efficient. On the other hand, building dams and reservoirs is very efficient and advantageous to the economy of the country. It attracts tourists, irrigates land, provides hydro-electric power and stores water for domestic use by the population of the country. However, it is also very expensive to build a dam or a reservoir, during the construction people have to be moved, diseases are prone from the water and highly depends whether the region has a source of water like river flowing. Lime Disease   Ã‚  Ã‚  Ã‚  Ã‚  Lyme disease is caused by Borrelia burgdorferi, which is a tick-borne spirochete. The dangers of this disease became more publicised in 1977, where a geographic grouping of children in Lyme, Conneticut were thought to have juvenile rheumatoid arthritis1. Soon after, it was discovered that lyme disease was an illness that mainly affects the skin, nervous system, heart, and joints. The borrelia species is part of the eubacterial phylum of spirochetes. Containted within a protoplasmic cylinder is a cell membrane, followed by wavy flagella, and then an outer membrane. The genes encoded within the outer membrane are located on plasmids which allows the organism to make antigenic changes in these proteins. When a borrelia cell attaches to its host, the whole outer membrane moves to one end of the cylinder, which is called capping to patching1. B. burgdorferi do not live in water, soil, or plants. Borrelia grow slowly compared to most bacteria. They elongate for 12 to 24 hours before dividing into two cells. B. burgdorferi is approximately 20 to um long and 0.2 to 0.25 um wide, with 7 to 11 flagella. More than 30 proteins are contained within B. burgdorferi1. This bacteria uses white-footed mice, mosquitoes, and deer as their hosts.   Ã‚  Ã‚  Ã‚  Ã‚  This disease does not discriminate between sex and age; male and female, as well as old and young are affected. It is widely distributed around the world in the temperate zones3. A person is infected when a black-legged tick imbeds itself into them while out in the open in wodded and forested areas. This usually occurs between the months of May and July. Tick abundance is associated with humidity, temperature, landscape slope, forested areas with sandy soils, and the extremity of residential development?.   Ã‚  Ã‚  Ã‚  Ã‚  Generally, lyme disease occurs in stages, which are not always clear-cut; they may overlap. The first stage involves the injection of B. burgdorferi by the tick. Shortly thereafter (3 days to 4 months), it spreads throughout the skin, causing erythema migrans (EM), which is basically a skin lesion. This lesion can vary in size, body site, color, duration, intensity, and recurrence. Erythema migrans is a marker of the disease, yet may also be absent altogether. EM resolves spontaneously in a few weeks or months4. Also like to occu... ...itive skills in children who have been treated for lyme disease using antibiotics. It seems likely that the lyme disease spirochete can cause an adverse fetal outcome. However, the question is, how likely and just what are the outcomes, which is what I would like to test for. My proposed study will be an experimental study in which lyme disease treated pediatric populations will be examined to identify possible cognitive or psychologic abnormalities resulting from lyme disease. The focus will be on children because they have a high incidence of lyme disease? and are less likely to have cognitive deterioration due to confounding factors, such as aging.   Ã‚  Ã‚  Ã‚  Ã‚  Children between the ages of 5 and 15 who have been treated with lyme disease will be studied. These children will be randomly chosen for endemic areas such as Delaware. Serologic testing (ie. enzyme-linked immunosorbent assa; ELISA) will be used to determine the presence of B. burgdorferi antibodies. The following hypotheses will be tested: Ho: No cognitive differences between lyme disease children and control group HA: Cognitive differences between lyme disease children and control group are present.

Romanic languages, group of languages belonging to the Italic subfamily of the Indo-European family of languages. They are spoken by about 670 million people in many parts of the world. Among the more important Romanic languages are Catalan, French, Italian, Portuguese, Romanian, and Spanish. My report is particularly about Italian and Spanish borrowings to the English language. Spanish borrowings appeared in English in 16 century. So the close cooperation with Spain contributed to the developing of borrowings from Spanish into English language. The force (weight) of air per unit area acting on object on the earth is called the atmospheric pressure (Diane, 11). The atmospheric pressure is measured by an instrument called a barometer. On a map, regions of equal pressure are linked by lines called isobars. These lines are drawn in such a way that they never cross each other. The drawing of these lines is based on the average sea level pressure reports. It is true that majority of the weather stations are situated some heath above sea level. Therefore the pressure measured at a particular place has to be converted into sea level pressure before the isobars are drawn. This is due to the fact that pressure decreases with increase in height and the pressures on maps should be as a result of weather conditions and not due to changes in height(Integrated Publishing). According to Diane, Pressure differences occur due to differences in the heat absorbed by the land or water. When air is heated, it expands reducing its density and rises. A partial vacuum is therefore created. Cold denser air flows to take up the space left by the rising air. Basically wind flows from a region of high pressure to a region of low pressure. Therefore, as the pressure changes from place to place, the speed of wind also changes. Diane defines pressure gradient as the rate of change of pressure at a particular time in a particular place. The force that pushes air from a region of high pressures to the region of low pressure is called the pressure gradient force. The force is greater when the pressure gradient is high. Eady notes that the speed of wind depends on the pressure gradient, being higher with greater pressure gradient. It could be expected that the wind will flow perpendicular to the isobars. However the pressure gradient force is somewhat circular as it depends not only on pressure gradient but also on Coriolis Effect, centrifugal force and frictional force (Eady, 142). Coriolis force is brought about as a result of the rotation of the earth. All particles at the surface of the earth rotate once every day. Since the radious is greater at the equator than towards the poles, an object rotates faster at the equator than at higher latitudes. If air moves northwards due to pressure gradient force, it maintains it horizontal motion; in accordance to Newton’s first law of motion (Ferrel, 98). The air therefore moves in a North Easterly direction. In actual sense, wind flows in a circular path (integrated Publishing). For a body to maintain a circular path, centripetal force must be provided. In accordance to Newton’s third law of motion an equal and opposites force called centrifugal force acts on the body (Eady, 139). This force tends to push winds out of their paths and it increases with speed of wind. As wind flows over objects over the earth’s surface it experiences fractional drag. This drag results to wind flowing at an angle to the isobars near the earth’s surface. At higher altitudes air flows parallel to the isobars (Joseph). Diane Fisher contends that in some cases, air cools at a particular place in the troposphere. The cooling results into contraction of air which in turn leads to increase in mass per unit volume of air. This leads to increased pressure directed downwards. This air then attempts to flow outwards towards regions of low pressure. But due to Coriolis force, the air is made to rotate clockwise in the northern hemisphere and anticlockwise in the southern hemisphere. This is referred to as an anticyclone. On the other hand, Diane tells us that a region of rising air of low pressure is called a cyclone. This occurs at the boundary of cold and warm air. It is caused by warm air rising and cools air descending which creates of reduced pressure. An inward pressure gradient is created and in addition to Coriolis Effect causes air to circulate in anticlockwise direction in the Northern Hemisphere and Clockwise in the Southern Hemisphere. The Pathogenesis of Atherosclerosis According to the World Health Organization, cardiovascular diseases are the leading cause of death worldwide, accounting for approximately 30 of all global deaths in 2008. A large proportion of CVDs is attributable to atherosclerosis, which is a major cause of myocardial infarction or stroke (1). The pathogenesis of atherosclerosis Over the past two decades, the inflammatory hypothesis of atherosclerosis has gained strong footing through multiple lines of supportive evidence (reviewed in (2)). Nowadays, atherosclerosis is considered a complex chronic inflammatory disease of medium- and large-sized arteries. Atherosclerosis occurs predominately at sites of disturbed laminar flow, in particular, arterial branch points and bifurcations. Human and animal studies indicate that the key initiating step is subendothelial accumulation of apolipoprotein B-containing lipoproteins (apoB-LPs). ApoB-LPs are secreted by the liver as very low-density lipoproteins, which are converted in the circulation to atherogenic low-density lipoproteins (LDL). In addition, apoB-LPs are secreted by the intestines as chylomicrons, which are converted by lipolysis into atherogenic particles, called remnant lipoproteins (3). Subendothelial apoB-LPs are believed to initiate an early inflammatory response, which may be enhanced by oxidative modification of LPs, through activation of overlying endothelial cells in a manner that leads to the recruitment of monocytes. Activated endothelial cells express adhesion molecules (e.g. intracellular adhesion molecule-1 and vascular adhesion molecule-1) and secrete cytokines and chemoattractants, or chemokines (e.g. monocyte chemoattractant protein-1 and RANTES), that act on monocytes and promote directional migration towards and into the artery wall. Once resident in the arterial intima, monocytes acquire the morphological characteristics of macrophages and increase their expression of scavenger receptors, including scavenger receptor A and CD36. Excessive uptake and internalization of modified lipoproteins via their scavenger receptors leads to the accumulation of cholesteryl esters in cytoplasmic droplets. These lipid-laden macrophages, known as foam cells, characterize the early atherosclerotic lesion (figure 1). As the atherosclerotic lesions further develops, macrophage and foam cells predominate, and further serve to alter the plaque environment, changing extracellular matrix composition and decreasing smooth muscle cell content, predisposing to plaque rupture (2, 4). Atherosclerosis and high-density lipoprotein therapy Although the development of atherosclerosis is dependent on a complex interplay between many factors and processes, a clear association has been established between elevated levels of plasma cholesterol and increased atherosclerotic disease (6). To attenuate the risk of atherosclerotic complications, primary and secondary prevention strategies seek to correct aberrant blood cholesterol levels. Actively reducing low-density lipoprotein (LDL) cholesterol through lipid-modifying therapies, such as statins, yield a proportional decrease in CVD risk (7). However, despite their potency, only 25-50% of cardiovascular events are prevented with highly potent statins, which highlights the importance of seeking for additional treatments for the optimal management of cardiovascular risk (8). Besides reducing LDL, improving HDL levels has gained a considerable amount of attention during the last decade. Epidemiological studies have shown that plasma levels of high-density lipoproteins (HDL) are inversely associated with clinical events resulting from atherosclerosis (9). Human and animal intervention studies have shown that increasing HDL results in a reduced atherosclerotic plaque size, suggesting that HDL may be an effective therapy for the regression of atherosclerosis (10). The mechanisms for plaque regression have been primarily attributed to the ability of HDL to promote cholesterol efflux from peripheral tissues, including macrophages, to the liver for excretion in the bile and feces. This process, called reverse cholesterol transport, is widely believed to account for much of the inverse relationship between plasma HDL levels and CVD revealed by population studies (11). HDL components can remove cellular cholesterol by four distinct processes. The presumed major precursor for this pathway is lipid-poor apoA-I, which is initially synthesized and secreted by the liver. Once in plasma, it rapidly acquires phospholipids and cholesterol from cell membranes in a reaction mediated by the ATP-binding cassette A1 (ABCA1) that results in the formation of pre-ÃŽÂ ² HDL particles (figure 2). A second mechanism involves ATP-binding cassette G1 (ABCG1), with pre-ÃŽÂ ² and large spherical HDL acting as the main acceptor. A third involves scavenger receptor B1 (SR-B1), which has the same acceptors as ABCG1. Lastly, cholesterol can be removed from cell membranes to HDL particles through passive diffusion. The latter three mechanisms are dependent on the presence and activity of Lecithin:Cholesterol AcylTransferase (LCAT). LCAT can esterify any unesterified cholesterol entering the outer surface of HDL, after which it will move into the intensely hydrophobic ce ntral core, leaving the outer surface of the HDL particle able to accept more unesterified cholesterol (12). Next to promoting cholesterol efflux, studies have shown that HDL is able to protect against cardiovascular diseases through a variety of additional functions, including anti-oxidant, anti-thrombotic, anti-apoptotic (reviewed in (12)). HDL has been shown to exert anti-inflammatory effects, which are mainly investigated in endothelial cells, and to a lesser extent in vascular smooth muscle cells (13, 14). Macrophages in the pathology of atherosclerosis It is generally accepted that macrophages play a pivotal role in the pathophysiology of atherosclerosis. The accumulation of macrophages and their conversion into foam cells, through the uptake of excessive amounts of lipids and cholesterol from modified apoB-LP, are considered hallmarks of atherogenesis. By expressing various effector molecules, including inflammatory cytokines, chemokines and extracellular matrix degrading enzymes, macrophages have a great impact on the activation, migration and survival of other cells in the plaque and ultimately affect plaque stability. However, within atherosclerotic plaques, macrophages represent a heterogeneous cell population, which may consist of several subsets that have distinct phenotypic and functional characteristics, ranging from large quiescent lipid-laden foam cells to a small active inflammatory cell. Furthermore, macrophages demonstrate a high degree of plasticity, which depend on the environmental cues they are exposed to. In gene ral, macrophages are skewed by interferon-ÃŽÂ ³ or lipopolysaccharide towards a pro-inflammatory or M1 phenotype, which produce mediators that have a pro-atherogenic effect. On the other hand, anti-inflammatory or M2 macrophages are polarized by interleukin (IL)-4, IL-10 and IL-13, which are believed to be of an anti-atherogenic nature. A phenotypical distinction can be made between these subsets based on their differential expression of cell surface expression (15, 16). High-density lipoproteins in modulating macrophages To date, HDL is considered to be the good cholesterol, because of its protective effects in atherosclerosis, such as anti-inflammatory properties (14). However, the effects of HDL on macrophages, which are major players in atherosclerosis, have yet to be established. The majority of the research conducted on the effects of HDL on macrophages have mainly been performed on cholesterol- or lipid-loaded macrophages. Here, HDL exerts anti-inflammatory effects by decreasing NF-ÃŽÂ ºB activation and secretion of pro-inflammatory cytokine, including tumor necrosis factor ÃŽÂ ± (TNFÃŽÂ ±), while increasing anti-inflammatory cytokines, like IL-10. To date, however, it is not known how HDL affects non-cholesterol or -lipid-loaded macrophages. ADAM 10 and 17 are closely related proteases and share many substrates, including TNF-ÃŽÂ ± and its receptor. ADAM activity can be regulated at various levels, including localization within the plasma membrane, where lipid rafts are thought to play a role in. Lipid rafts are cholesterol- and sphingolipid-enriched microdomains within the cell membrane, which are able to include or exclude proteins to variable extents. This dynamic process regulates protein interactions and influences their functions. Lipid rafts can be decreased and disrupted by cholesterol depletion, e.g. by HDL (18). Tellier et al. showed in vitro that TNFÃŽÂ ± shedding was increased in fibroblasts and ECs after incubation with HDL. This was attributed to an increased activity of ADAM 17, which was due to lipid raft disruption by cholesterol depletion (19). Study aim and design The purpose of this study is to investigate the effects of HDL on macrophage phenotype and whether NF-kB signaling, lipid raft disruption and increased activity of ADAM10 and 17 are involved in this. First, we will determine the effects of HDL on M1 and M2 macrophage polarization by exposing bone marrow-derived macrophages (BMDMs) from C57BL/6 mice to HDL. Here, M1 and M2 polarization markers will be determined using quantitative PCR and ELISA. Second, we will examine whether NF-ÃŽÂ ºB signaling is involved in the pro-inflammatory effects induced by HDL in macrophages.
